Pasquini was born in Ferrara. He studied with Alessandro Milleville (1521-1589).
On May 1, 1592 Pasquini became organist of the ''Ridotti of Mario Bevilacqua'' and of the ''Olivetian Church of Santa Maria in Organo'' in Verona.
On October 6, 1597 he was elected organist of the ''Capella Giulia'' in St. Peter's Basilica in Rome. In the summer and autumn of 1604 he took over the same position at the ''Santo Spirito'' in Sassia and continued his work at St. Peter's.
Based on a report by Agostino Faustini in 1646, Pasquini presumably died in Rome.
